Traditional Use: Direct Substrate Inoculation (Spawn Replacement)
The traditional way to use Preta Pearls: As a drop-in, grain-free alternative to traditional spawn.
Steps: After forming and rinsing pearls (per the making tutorial), add them directly to pasteurized or supplemented substrates like sawdust, straw or coco coir.
Inoculation Rates: Base it on LC equivalents for efficiency. If you’d normally use 5cc LC per kg dry substrate (standard for oysters), start with 5cc worth of pearls. But since they’re protected and contamination-resistant, safely ramp up to 20cc beads per kg for accelerated colonization.
Benefits: Pearls disperse evenly for multiple inoculation points, leading to visible mycelium clouds in 4–7 days. No endospores or clumping like grain; the alginate shell protects while allowing emergence.
Tips:
- Add 0.5–2% gypsum to the substrate for calcium buffering.
- Mix gently to avoid crushing beads. But if got calcium in substrate then will spherite broken beads.
- We use BioaKt as supplement for Ca+ ion source.

Expect radial rhizomorphic growth bridging beads quickly as show in image.
This method cuts out spawn-run wait times, 2-3 weeks if all well, making it perfect for hobbyists or small farms. The downsides are:
- Volumes required – but can easily augmented by using alginate spawn as G1 and Supplemented sawdust as G2.
- Flowhood – Even if we have eliminated a lot with Selective Nutrients (VermiLIN), full aSeptic methodologies still required and imperative for supplemented substrates that are sterilized.
Advanced Technique: In-Situ Pearl Formation (Direct Injection into Substrate)
Take contamination risks down to near-zero by forming pearls inside the substrate—no handling wet beads at all.
- Steps: Load a syringe with your LC + activator mix (undripped, per making tut). Inject directly into a prepared, calcium-boosted substrate (e.g., PF-Tek jars, CVG bags or BioaKt supplemented logs).
- How It Works: The substrate’s calcium ions (from added gypsum, CaOH, CaCO3, etc) trigger spherification—alginate gels into pearls in-situ, solidifying without added water issues.
- Rates & Setup: Use LC-equivalent doses (5–20cc mix per kg dry sub). Supplement with 0.5–6% gypsum (e.g., 6% in CVG for strong cross-linking) or use BioaKt – 1% Calcium ion source required.
- Benefits: Skips the entire extract/rinse/apply cycle, slashing contamination vectors (no open-air exposure, SHIP→ Syringe→ Substrate). Pearls form and emerge locally for radial spread—”set it and forget it” inoculation.
- Tips: Opt for a larger-gauge needle if the mix is viscous, we find 16G does job well. Test on small batches first; great for scale-up in contam-prone spaces. Mycelium activates naturally as pearls stabilize.
This innovation makes alginate even more user-friendly, especially for beginners avoiding SAB work.
Long-Term Storage: Pearls as Bundu Castellani Upgrade
For vaulting cultures without fridges or repeated transfers, Preta Pearls shine in stasis.

- Steps: Form pearls as usual, extract from the bath (no rinse needed for storage), and add aseptically to sterile distilled water (0.0 EC) in vials or jars.
- Why It Works: Instant Bundu Castellani-style preservation—but enhanced. The alginate bead replaces the agar wedge, with carbon adding Terra Preta longevity for microbial stability.
- Extension to Long-Term LC: Beads stay “awake” briefly post-formation, growing mycelium into the surrounding water (creating cloudy, viable suspensions—see photos of myceliated water). This evolves into a stable, extractable “long-term LC”: Pull the mycelium-filled water as fresh inoculum. Better than traditional Castellani’s “shake to activate”—no agitation required, just natural revival.
- Viability & Tips: Expect 1–5+ years (test by plating small samples periodically). Optional: Add trace VermiLIN (30ml/L) for subtle support without triggering full growth. Always use a SAB or flow hood for transfers to avoid rare bacterial intrusion.
- Revival: Drop beads straight into substrate, or use the myceliated water as LC for quick starts.
- TIP: for ease of use and sterility use a SHIP culture bottle.
